I was wondering if there is a way to update the definition file for Malwarebytes Anti-Malware for a PC that is offline, without access to the internet?

There is. You can use this site to download the file, or, you could manually do this yourself if you have access to a machine with internet access. The file is called rules.ref and is located in different locations depending on your Windows version.

For Windows XP it's normally under; Documents and Settings\All Users\Application Data\Malwarebytes\Malwarebytes' Anti-Malware\>

For Windows 7 it's normally under; ProgramData\Malwarebytes\Malwarebytes' Anti-Malware\>
